    Background
    LIMP-2 (lysosomal integral membrane protein type 2)/SCARB2, is a lysosomal membrane glycoprotein and the only endomembrane member of the CD36 scavenger receptor family, which also includes SR-B1 and CD36. Structurally, it contains two transmembrane domains, short cytosolic tails, and a large luminal ectodomain with a hydrophobic tunnel-like cavity that binds and transports cholesterol. Highly expressed in lysosomes, LIMP-2 serves multiple functions: it delivers the lysosomal enzyme β-glucocerebrosidase, acts as a receptor for certain enteroviruses, and mediates lysosomal cholesterol export in parallel with NPC1/2. By shuttling cholesterol (and potentially other lipids) from the lysosomal lumen to the limiting membrane and lipid droplets, LIMP-2 regulates cholesterol homeostasis, LDL receptor expression, and SREBP-2 signaling, linking it to lipid metabolism and lysosomal storage disorders.
