    Background
    RAD52 is a human DNA/RNA-binding protein of 418 amino acids that forms multimeric, ring-shaped oligomers—historically described as heptamers with its highly conserved N-terminal domain (aa 1–208) mediating DNA binding and oligomerization, and its less conserved C-terminal domain containing binding sites for RAD51 and replication protein A (RPA). RAD52 is expressed in the nucleus of proliferating cells, where it plays multifaceted roles in DNA double-strand break repair pathways, most prominently in single-strand annealing (SSA) and as a backup factor for homologous recombination (HR) in BRCA-deficient contexts. In SSA, RAD52 anneals complementary single-stranded DNA overhangs and stimulates ERCC1–XPF nuclease activity, while in HR it can facilitate RAD51 loading, promote second-end capture, and maintain genomic stability. Although dispensable for viability in normal vertebrate cells, RAD52 becomes essential under specific DNA repair deficiencies, making it an attractive target for synthetic lethality-based cancer therapies.
