    Background
    The Foxd3 gene encodes a transcriptional repressor that belongs to the Forkhead Box (Fox) family, a group of proteins defined by a conserved winged-helix DNA-binding domain of approximately 100 amino acids, along with regions that mediate transcriptional activation or repression. Initially identified for its expression in embryonic stem (ES) cells and multipotent neural crest (NC) cells, FoxD3 plays a critical role in early development. As a key transcriptional regulator, FoxD3 is essential for maintaining pluripotency during the pre-implantation and peri-implantation stages of mouse embryogenesis and is also necessary for proper trophoblast development. Beyond its role in early embryonic stages, FoxD3 is crucial for the development and maintenance of the mammalian neural crest. Additionally, FoxD3 participates in a core regulatory network with other key pluripotency factors such as Oct-4 and NANOG, working together to preserve the undifferentiated state of ES cells and regulate cell fate decisions during development.
