    Background
    Histone deacetylases (HDACs) are enzymes that remove acetyl groups from lysine residues on both histone and non-histone proteins. They are central to transcriptional regulation and are implicated in nearly all chromatin-associated biological processes in eukaryotes. The human class I HDACs, which share homology with the yeast RPD3 protein, include HDAC1, HDAC2, HDAC3, and HDAC8. Among these, HDAC3 functions as a regulator of gene expression by deacetylating histones as well as numerous non-histone substrates. Its activity is tightly controlled through interactions with the corepressors N-CoR and SMRT, forming a functional complex that mediates transcriptional repression. HDAC3 localizes to both the nucleus and cytoplasm and targets a broad range of substrates, including histones H2A, H3, and H4, and non-histone proteins such as RelA, SRY, p53, MEF2, PCAF, and p300/CBP. The HDAC3–N-CoR–SMRT complex is essential for silencing transcription driven by nuclear hormone receptors and other transcription factors, including Rev-Erb, COUP-TF, DAX1, MAD, and Pit-1. Furthermore, HDAC3 enzymatic activity is enhanced by phosphorylation at Ser424 by casein kinase 2 (CK2), while dephosphorylation by protein phosphatase 4 (PP4) reduces its activity.
