    Background
    Thymidylate synthase (TS) catalyzes the reductive methylation of deoxyuridine monophosphate (dUMP) to thymidylate (TMP), using 5,10-methylenetetrahydrofolate as the methyl donor. This reaction represents the sole de novo pathway for intracellular TMP production, a nucleotide that is indispensable for DNA synthesis and repair. Inhibition of TS results in thymidylate depletion, triggering thymineless death—a process marked by extensive DNA damage, chromosomal abnormalities, and apoptosis. The anticancer drug 5-fluorouracil (5-FU) functions as a TS inhibitor and has demonstrated efficacy against various solid tumors, including those of the colon, breast, head, and neck. Clinical studies have shown that tumors with low TS expression are more responsive to 5-FU therapy compared to those with high TS levels. Beyond its role as a therapeutic target, TS is essential for cell proliferation and survival, and its expression has been associated with multiple cancer types.
