    Background
    Desmoplakin I+II are major high–molecular weight proteins of the desmosomal plaque that function as essential linkers between intermediate filaments (IFs) and cell–cell junctions, thereby providing tissue integrity under mechanical stress. Both isoforms are encoded by the same gene through alternative splicing, with DPI containing a long central α-helical coiled-coil rod domain (~132 nm) and DPII having a shorter rod due to deletion of 599 residues. Their structure is organized into three regions: an N-terminal domain mediating desmosome targeting, a central rod promoting dimerization, and a C-terminal domain with repeated motifs that directly interact with IFs such as keratins, vimentin, and desmin. DPs are highly expressed in desmosome-rich tissues such as epidermis and myocardium, where they stabilize intercellular junctions by anchoring the IF cytoskeleton to the plasma membrane. Functionally, they are crucial for maintaining epithelial and cardiac tissue integrity, and mutations in DP cause skin blistering disorders and cardiomyopathies due to defective IF attachment and desmosomal stability.
