    Background
    Jagged2 is a canonical mammalian Notch ligand belonging to the Serrate/Jagged family, characterized by an N-terminal C2 lipid-binding domain, a receptor-binding DSL domain, and multiple EGF repeats that mediate interactions with Notch receptors. Structurally, Jagged2 shares homology with Jagged1 but differs in its signaling strength and sensitivity to glycosylation modifications such as Fringe-mediated GlcNAc addition. It is expressed in various embryonic and adult tissues, particularly at sites of epithelial–mesenchymal interactions and within the hematopoietic niche. Functionally, Jagged2 activates Notch1 and Notch2, promoting T-cell lineage differentiation while inhibiting B-cell and myeloid development, thereby contributing to developmental processes and fine-tuning hematopoietic lineage outcomes through ligand-specific Notch signaling strength and receptor selectivity.
