Method

AML cell lines are seeded in triplicate in 96-well plates (5–9 × 103cells/well) ± doxycycline (100 ng/mL). Viable cells are measured using CellTiter 96 AQueous One Solution MTS Reagent for Cell Proliferation Assays. For Colony-Forming Assays, AML Cell Lines are seeded in duplicate at 0.1–0.2 × 103/dish in MethoCult H4230 ± 100 ng/mL doxycycline or in the presence and absence of graded concentrations of NRD167. Colonies are counted 7 to 10 days after planting.
