  • Cell lines: CD34+ HSPCs, iPSCs and fibroblasts
  • Concentrations: 1 μM, 5 μM, 100 μM
  • Incubation Time: 72 h, 5 days, 14 days
  • Method:

    Equal numbers of DC patient iPSCs are plated and 24 hr later treated with 1 μM BCH001 versus DMSO as vehicle control. 72 hr post-treatment cells are harvested and fixed using 70% ethanol at 4 ℃ overnight.
    For CD34+ HSPCs, 1000 cells are plated in methylcellulose medium containing human cytokines and inhibitors BCH001, RG7834 or vehicle (DMSO). After 14 days of culture at 37℃, 5% CO2, the progenitor colonies are counted manually under the microscope, with categories defined by colony morphology.
