Method

Sub-confluent MDA-MB-468 cells were serum-starved in DMEM/0.1% BSA for 18 h, pretreated overnight with SPI-112 (100 μM) and stimulated with EGF (50 ng/ml, 10 min) or mocked treated as indicated. Shp2 was immunoprecipitated from cell lysate supernatants and Shp2 PTP activity was measured.
