PH domain and leucine rich repeat phosphatase (Phlpp) inhibitor NSC45586 increases Pth1r mRNA and protein expression in ATDC5 cells and IMCs within 24 hours. [2]

Method

ATDC5 cells were seeded at a density of 5 x 105 cells/well in a six-well plate and incubated overnight to allow for cell adhesion in DMEM supplemented with 10% FBS and 100 units/mL penicillin, 100 μg/mL streptomycin, and 0.25 μg/mL Amphotericin B (1% antibiotic/antimycotic). Phlppi including NSC45586 (25 μM) treatments were added. Primary chondrocytes were plated and allowed to adhere overnight as described above. Actinomycin D (5μM) was added for six hours and then replaced with medium containing Phlppi including NSC45586 or vehicle for 24 hours. Cells were collected for RNA or protein extraction.
