Description NSC45586 is an inhibitor of PHLPP (Pleckstrin homology domain and leucine-rich repeat protein phosphatase), which is selective for PHLPP1 and PHLPP2.

    ATDC5 cells were seeded at a density of 5 x 105 cells/well in a six-well plate and incubated overnight to allow for cell adhesion in DMEM supplemented with 10% FBS and 100 units/mL penicillin, 100 μg/mL streptomycin, and 0.25 μg/mL Amphotericin B (1% antibiotic/antimycotic). Phlppi including NSC45586 (25 μM) treatments were added. Primary chondrocytes were plated and allowed to adhere overnight as described above. Actinomycin D (5μM) was added for six hours and then replaced with medium containing Phlppi including this compound or vehicle for 24 hours. Cells were collected for RNA or protein extraction.
