Auristatin E (AE) is a fully synthetic, highly potent antimitotic pentapeptide and a structural analog of dolastatin-10. It inhibits microtubule polymerization, leading to mitotic arrest at the G₂/M phase and subsequent apoptotic cell death in tumor cells. Due to its high toxicity, AE is primarily utilized as a cytotoxic payload in antibody–drug conjugates (ADCs), where targeted delivery minimizes systemic toxicity.

Mechanism of Action

In vivo

Auristatin E (0.25 mg/kg, s.c., a single dose for 80 days) inhibits tumor growth in nude mice with subcutaneous L2987 human lung adenocarcinoma or 3396 human breast carcinoma xenografts.
