Description SSE15206 is a tubulin polymerization inhibitor that has potent antiproliferative activities in cancer cell lines of different origins and overcomes resistance to microtubule-targeting agents.

Treatment of cells with SSE15206 causes aberrant mitosis resulting in G2/M arrest due to incomplete spindle formation, a phenotype often associated with drugs that interfere with microtubule dynamics. SSE15206 inhibits microtubule polymerization both in biochemical and cellular assays by binding to colchicine site in tubulin. Prolonged treatment of cells with SSE15206 results in apoptotic cell death accompanied by p53 induction[1].

Incubation Time 4, 8, 12, 24 and 36 hours
Method

HCT116 cells are treated with 0.5 μM, 1 μM and 2 μM SSE15206 for 4 and 8 hours. Phosphorylation of histone H3 (S10) and P-MPM2 is determined by western blotting; total-histone H3 and GAPDH are used as loading controls; Or HCT116 cells are treated with 1 μM SSE15206 for 4, 8, 12, 24 and 36 hours and are analyzed through FACS following treatment.
