Biological Activity

Description RGD peptide (GRGDNP) is an inhibitor of binding of integrins to the extracellular matrixs. RGD peptide (GRGDNP) induces apoptosis presumably through direct activation of caspase-3.
Targets
integrin [1] caspase-3 [2]
In vitro

RGD-peptide interfers with cell proliferation by affecting cell protrusion and stress fibers.[3]

Protocol (from reference)

Cell Research:

[3]

  • Cell lines: MC3T3-E1: ATCC CRL-2594 cell line
  • Concentrations: 1,000 µM
  • Incubation Time: 24 h
  • Method:

For inhibition of cell attachment, MC3T3-E1 cells were incubated at 37°C for 24 hours in the presence of 5 mM EDTA, 1,000 µM RGD, 10 µg/mL MAB1998, 0.2 µL IgG, and 10 µg/mL heparin. Next, the cells (500 µL, 5,000 cells/well) were added to the wells and incubated for 24 hours. To measure cell proliferation on the specimens, a metabolic activity cell counting kit assay was used in this experiment.

Calculation results:

Working concentration: mg/ml;

Method for preparing DMSO master liquid: mg drug pre-dissolved in μL DMSO ( Master liquid concentration mg/mL, Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug. )

Method for preparing in vivo formulation: Take μL DMSO master liquid, next addμL PEG300, mix and clarify, next addμL Tween 80, mix and clarify, next add μL ddH2O, mix and clarify.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next add μL Corn oil, mix and clarify.

Note: 1. Please make sure the liquid is clear before adding the next solvent.
2. Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such
as vortex, ultrasound or hot water bath can be used to aid dissolving.
