It also acts as a competitive inhibitor of acyl peptide substrate, binding directly within the Sirt4 acyl-binding pocket and noncompetitive with the NAD+ cofactor. SIRT4-IN-1 treatment effectively increases glutamate dehydrogenase (GDH) activity and rescues pyruvate dehydrogenase (PDH) activity.
