JNJ-6640 is a first-in-class, selective inhibitor of Mycobacterium tuberculosis PurF(amidophosphoribosyltransferase), the initiating enzyme in de novo purine biosynthesis. It also exhibits potent in-vitro bactericidal activity against M. tuberculosis with an MIC90 of 8.6 nM and disrupts purine production, leading to impaired DNA replication and bacterial lysis.
