Cells were treated with sinomenine for 24 h. The expressions of S phase cell cycle-regulating proteins were measured by western blot analysis.
Int J Oncol, 2016, 48(5):2098-112. Sinomenine purchased from Selleck.
Repeated i.p administration of sinomenine reversed TCI-induced upregulation of JAK2/STAT3 pathway in the spinal cord. (a to d) Western blot data summary showing that sinomenine decreased the protein expression of p-JAK2 and p-STAT3 (***p < 0.001 compared with the sham + vehicle group; ###p < 0.001 compared with the CIBP + vehicle group, n = 6 in each group). Sinomenine (i.p, 40 mg/kg) or vehicle was administered once a day from day 14 after TCI to day 18. Tissues were collected 2 h after the last injection. GAPDH: glyceraldehyde-3-phosphate dehydrogenase antibody; CIBP: cancer-induced bone pain; p-JAK2: phosphorylated-Janus family tyrosine kinase 2; p-STAT: phosphorylated-signal transducers and activatorsof transcription 3.

SINO reduced neuronal loss and apoptosis after MCAO. A-B. Nissl staining and quantitative analysis of neuronal survival at 72 h after MCAO (n = 5). Scale bar = 100 μm. C–D. Western blot and quantitative analysis of cleaved caspase-3 expression in the ischemic hemisphere at 72 h after MCAO (n = 5). Data are means ± SEM, *P < 0.05 and **P < 0.01, vs. sham group; #P < 0.05 and ##P < 0.01 vs. the indicated groups. SINO-10: 10 mg/kg SINO; SINO-20: 20 mg/kg SINO.

Sinomenine, a pure alkaloid extracted from the chinese medical plant Sinomenium acutum, is used for the treatment of rheumatism and arthritis. However its analgesic action against other kinds of pain is limited. Sinomenine is a morphinan derivative, related to opioids such as levorphanol and the non-opioid cough suppressant dextromethorphan. Its anti-rheumatic effects are thought to be primarily mediated via release of histamine, but other effects such as inhibition of prostaglandin, leukotriene and nitric oxide synthesis may also be involved.

* Please note that Selleck tests the solubility of all compounds in-house, and the actual solubility may differ slightly from published values. This is normal and is due to slight batch-to-batch variations.

Method for preparing in vivo formulation：Take μL DMSO master liquid, next addμL PEG300， mix and clarify, next addμL Tween 80，mix and clarify, next add μL ddH2O，mix and clarify.
1.Please make sure the liquid is clear before adding the next solvent.
2.Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such as vortex, ultrasound or hot water bath can be used to aid dissolving.
Calculate the mass, volume or concentration required for a solution. The Selleck molarity calculator is based on the following equation:
Mass (mg) = Concentration (mM) × Volume (mL) × Molecular Weight (g/mol)
*When preparing stock solutions, please always use the batch-specific molecular weight of the product found on the via label and MSDS / COA (available on product pages).
Calculate the dilution required to prepare a stock solution. The Selleck dilution calculator is based on the following equation:
Concentration (start) x Volume (start) = Concentration (final) x Volume (final)
This equation is commonly abbreviated as: C1V1 = C2V2 ( Input Output )
* When preparing stock solutions always use the batch-specific molecular weight of the product found on the vial label and MSDS / COA (available online).

Definitions of molecular mass, molecular weight, molar mass and molar weight:
Molecular mass (molecular weight) is the mass of one molecule of a substance and is expressed in the unified atomic mass units (u). (1 u is equal to 1/12 the mass of one atom of carbon-12)
Molar mass (molar weight) is the mass of one mole of a substance and is expressed in g/mol.
