  • Cell lines: HCT116 and SW480 cells
  • Concentrations: 20 μM
  • Incubation Time: 2 h
  • Method:

    HCT116 and SW480 cells (as indicated) were pretreated with the inhibitors Z-IETD-FMK (caspase 8 inhibitor), Z-LEHD-FMK (caspase 9 inhibitor), Z-VDAVD-FMK (caspase 2 inhibitor), and Z-AEVD-FMK (caspase 10 inhibitor; 20 μm each) for 2 h, after which TRAIL (50 ng/ml) was added. After 4 h (or 16 h for caspase 3 activation) of TRAIL exposure, extracts were collected and analyzed by Western blotting for the expression of the different proteins.
