In vitro

SN-011 substantially suppresses 2′3′-cGAMP-induced STING oligomerization and phosphorylation assessed by immunoblot in HFFs. This compound specifically inhibits STING activation and STING-dependent signaling.

In vivo

Serum antinuclear antibody is markedly reduced by SN-011 treatment. This compound also significantly reduces the number of activated CD69+ CD8 T cells and memory CD44high CD62Llow CD4 and CD8 T cells to near normal levels in the spleens of treated Trex1−/− mice. It strongly reduces inflammation and protects Trex1−/− mice from death.
