Biological Activity

In vitro
In vitro The treatment of Jurkat T cells with thimerosal causes a significant decrease in cellular viability at 1 μM (25%, p<0.05; IC50: 10 μM)[1]. Thimerosal is also able to induce apoptosis and G2/M phase in human leukemia U937 cells[2].

Method The number of 1×104 cells/well Jurkat T cells are seeded into 96 well plates and exposed to thimerosal, methyl mercury, and mercuric chloride (HgCl2) at concentrations of thimerosal (0.01-0.1-1-10-50-100-250 μM), methyl mercury (30-50-80-100-250 μM), and mercuric chloride (HgCl2 (20-40-60-80-100 μM). The plates are incubated at 37°C in a humidified atmosphere containing 5% CO2. After 48 hours, the medium is discarded and 20 μl/well of MTT solution (5 mg/ml) is added and incubated for 3 hours at 37°C (5% CO2). Finally, 20μl/well of isopropanol is added and the color intensity is read spectrophotometrically at 590 nm.
