Method

Immortalized mouse renal MCs (SV40-MES-13) were cultured in DMEM/F12 containing 5% fetal bovine serum (FBS), 0.5% penicillin and streptomycin in an atmosphere of 5% CO2 at 37 ℃. Following overnight serum deprivation, the SV40-MES-13 cells are seeded on cover slips coated with either 2 kPa (soft) or 50 kPa (stiff) hydrogels, and then treated with the YAP inhibitor Super-TDU, Piezo1 activator Yoda-1, MEK1/2 inhibitor PD0325901, JNK inhibitor SP600125, or the p38 kinase inhibitor SB203580 for 48 h.
