Biological Activity

Description Tectoridin (Shekanin), an isoflavone isolated from flowers of Pueraria thunbergiana, has several pharmacological effects including anti-cancer, anti-inflammatory, antioxidant, and hepatoprotectivy.
In vitro

Tectoridin is known as a phytoestrogen. Tectoridin exerts its estrogenic effect mainly via the GPR30 and ERK-mediated rapid nongenomic estrogen signaling pathway. It bound poorly to estrogen receptor α. Tectoridin increased intracelluar cAMP accumulation and rapidly increased the phosphorylation of ERK1/2, but does not induce phosphorylation of ER α at Ser118[1].

In vivo Tectoridin shows significant protective effect on hepatic steatosis induced by ethanol through the modulation of PPAR pathway and protection of mitochondrial injury[2].

  • Cell lines: MCF-7 human breast cancer cells
  • Concentrations: 0-10 μM
  • Incubation Time: 96 h
  • Method:

    MCF-7 cells (5 × 105) are seeded on 60 mm dishes with hormone-free experimental medium and incubated for 48 h at 37°C in order to synchronize the cells in the G0/G1 phase. The cells are then treated with experimental medium supplemented with various concentrations (0-10 μM) of genistein, tectorigenin and tectoridin for 96 h. The cell cycle was analyzed.
