EG1, a specific and efficacious Pax2 inhibitor that phenocopies a genetic loss of function, is capable of inhibiting Pax2 mediated expression of EGFP without significant effects on Pax2 protein levels.[1]

Method

PRS4-EGFP cells transfected with CMV-Pax2b or CMV-Pax2bC63Y (Mut) and treated with increasing concentrations of EG1. The media were removed from test plates, and cells were lysed in 2×SDS buffer. Total protein was separated on 4-12% Bis-Tris gels, transferred to PVDF membranes, and immunoblotted with anti-Pax2 antibody,20 anti-EGFP antibody, antiactin, anti-P-Histone H3, anti-Histone H3, or anti-Actin.
