Benzoylaconine (Isaconitine, Pikraconitin) is an alkaloid derived from the Chinese traditional medicine Radix Aconiti Lateralis Preparata (Fuzi). Benzoylaconitine inhibits production of IL-6 and IL-8 via MAPK, Akt, NF-κB signaling in IL-1β-induced human synovial cells.

Benzoylaconitine is a lower toxic monoester type alkaloid considered as the pharmacodynamic constituent in Aconitum species, which exerts an anti-inflammatory effect dependent on MAPK, Akt and nuclear factor-κB (NF-κB) pathways in human synovial cells stimulated with IL-1β.[1]

Method

The SW982 cells are pretreated with various doses (0, 5 and 10 μM) of BAC for 30 min, followed by IL-1β stimulation (10 ng/mL) for 12 h. The levels of IL-6 and IL-8 in supernatants of cell cultures are determined by ELISA kits according to the manufacturer's recommendations.

Benzoylaconitine shows efficient in vivo anti-inflammatory effect with high ear (69.8 %) and paw (87.1 %) swelling suppressing rate.[3]
