In vitro

WST-8 is reduced by NADH in good yields at neutral pHs in the presence of 1-methoxy PMS to produce the corresponding formazan dye that absorbs at 460 nm. The formazan is soluble to water at concentrations higher than 0.1 M. WST-8 is useful as a sensitive chromogenic indicator for NADH. It is also applicable to cell proliferation assays as a cell viability indicator.[1]

Cell Research Cell lines Hela cells
Concentrations 0.5 mM
Incubation Time 2 h
Method

A hundred μl of HeLa cell culture in the logarithmic phase is inoculated onto a 96-well microtiter plate, and the plate pre-incubated for 24 h in a CO2 incubator at 37℃. A 10 μl of the working solution containing WST-8 and lmethoxy PMS (0.5 mM and 20 μM, respectively, as the final concentration) is added to each well; it is incubated for an additional 2 h. The absorbance of each well is measured at 450 nm with a reference wavelength at 650 nm with an M-UVmax microplate reader.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next addμL PEG300, mix and clarify, next addμL Tween 80, mix and clarify, next add μL ddH2O, mix and clarify.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next add μL Corn oil, mix and clarify.

Note: 1. Please make sure the liquid is clear before adding the next solvent.
2. Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such
as vortex, ultrasound or hot water bath can be used to aid dissolving.
