Description Rosavin, a cinnamyl alcohol glycoside found in the plant Rhodiola rosea, possesses antidepressant and anxiolytic activities.
In vivo Rosavin is eliminated in vivo quickly. T1/2z for i.v. is 2.1 ± 0.5 h. Tmax for p.o. is 0.9 ± 0.6 h, which indicates that the concentration of rosavin reached Cmax and it is fast absorbed in vivo ∼ 50 min after oral administration. Vz/F for i.v. is 17.6 ± 5.8 L/kg, which demonstrates that most of rosavin is distributed in extracellular fluid. CLz for p.o. is 93.8 ± 65.6 L/h/kg, which shows that rosavin experiences a slow elimination in vivo after oral administration. AUC(0−∞) for p.o. is only 850.1 ± 766.1 μg/L h, and Vz/F for p.o. is much larger than that of i.v., which may be caused by highly water-soluble, polar and lower oil-water partition coefficient of rosavin, leading to the difficult absorption of the drug in vivo. Furthermore, rosavin is one of the glycoside compounds, which will be quiet easily hydrolyzed to aglycone in an acidic environment. The oral bioavailability is 4.7%. It may result from the poor membrane permeability, significant first-pass metabolism and efflux vector of small intestine and so on[1].
