Trans-Zeatin

Synonyms: (E)-Zeatin

Trans-Zeatina ((E)-Zeatin) is the member of the plant growth hormone family known as cytokinins, which regulate cell division, development, and nutrient processing. Trans-Zeatin inhibits UVB-induced MMP-1 expression, c-Jun activation and phosphorylation of ERK, JNK and p38 MAP kinases (MAPKs) dose-dependently.

Biological Activity

Description Trans-Zeatina ((E)-Zeatin) is the member of the plant growth hormone family known as cytokinins, which regulate cell division, development, and nutrient processing. Trans-Zeatin inhibits UVB-induced MMP-1 expression, c-Jun activation and phosphorylation of ERK, JNK and p38 MAP kinases (MAPKs) dose-dependently.
Targets
MMP-1 [1] ERK [1] JNK [1] p38 MAPK [1]
In vitro
In vitro Pretreatment with trans-Zeatin significantly inhibits UVB-induced MMP-1 expression and c-Jun activation in a dose-dependent manner in cultured human skin fibroblasts, which may be mediated by inhibition of ERK, JNK and p38 MAPKs signaling pathways[1].
Cell Research Cell lines human skin fibroblasts (HSFs)
Concentrations 20, 40, 80, 150, 300, 600 μM
Incubation Time 24 h
Method HSFs are seeded in a 96-well plate. After various concentrations of trans-Zeatin or UVB treatment, cells are cultured in 100 μl 10% DMEM in each well for the indicated time. MTT solution (10 μl) at a concentration of 5 mg/ml is added to each well, and the cells are then incubated at 37˚C for 4 h. The medium is removed, and 200 μl of DMSO is added into each well to dissolve formazan crystals by pipetting up and down several times. The absorbance is measured spectrophotometrically on an ELISA plate reader at a wavelength of 570 nm.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next addμL PEG300, mix and clarify, next addμL Tween 80, mix and clarify, next add μL ddH2O, mix and clarify.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next add μL Corn oil, mix and clarify.

Note: 1. Please make sure the liquid is clear before adding the next solvent.
2. Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such
as vortex, ultrasound or hot water bath can be used to aid dissolving.
