In vitro 1,2,3,4,5,6-Hexabromocyclohexane inhibits Jak2 autophosphorylation in a time- and dose-dependent manner. 1,2,3,4,5,6-Hexabromocyclohexane is noncytotoxic to cells at concentrations that maximally inhibit Jak2 tyrosine autophosphorylation[1].

Method

BSC-40 cells are transfected/infected. Prior to cell lysis, 50 µM 1,2,3,4,5,6-hexabromocyclohexane is applied to the cells for 0, 1, 4, or 16 h. Cellular lysates are then prepared, immunoprecipitated with anti-Jak2 antibody, and immunoblotted with anti-phosphotyrosine antibody to measure tyrosine phosphorylated Jak2 levels.
