Biological Activity

Description Kartogenin (KGN) is an activator of the smad4/smad5 pathway, and promotes the selective differentiation of multipotent mesenchymal stem cells into chondrocytes.

In vitro

Kartogenin induces chondrocyte differentiation from hMSCs by enhancing the nuclear localization of CBFβ, and protects articular chondrocytes. [1] Kartogenin promotes the type-I collagen synthesis of dermal fibroblasts via activation of the smad4/smad5 pathway without obvious cytotoxicity on fibroblasts’ viability, morphology and survival. [2]

In vivo Kartogenin promotes cartilage repair in collagenase VII (10 μM)– and surgery (100 μM)-induced OA models in mice. [1] Kartogenin (100 nM) also stimulats collagen synthesis in the mouse dermis. [2] In addition, Kartogenin stimulates mouse limb skeletal growth and synovial joint development. [3]

  • Cell lines: Fibroblasts
  • Concentrations: ~1 μM
  • Incubation Time: 48 hours
  • Method: For the cell viability assay, fibroblasts are seeded on six-well plates, followed by Kartogenin or DMSO treatment. According to the manufacturer’s instructions, cell viability and viability are assessed by AlamarBlue assay.
