Mechanism of Action

In vitro

Pramoxine is used primarily in combination with colloidal oatmeal, hydrocortisone, menthol, and petrolatum. [1]

In vivo

Pramoxine (1.7 mM) causes a lower degree of attenuation of the response to substance P than the responses to acetylcholine, histamine and barium chloride on the guinea-pig ileum. Pramoxine exhibits anesthetic inhibitory potency on the guinea-pig ileum with pKa of 6.4. [2] Pramoxine produces shifts in specific skin temperature at which activation and suppression of both panting and shivering occur in dogs, these changes are interpreted as indicating peripheral mediation in control of thermoregulatory end mechanisms. [3]
