Rapamycin (Sirolimus) is a specific mTOR inhibitor that complexes with FKBP12 to allosterically inhibit the mTORC1 complex. Disruption of mTORC1 downstream signaling leads to the suppression of protein synthesis, cell cycle progression, and cellular proliferation while potently triggering autophagy. Furthermore, rapamycin inhibits interleukin-mediated hematopoietic bone marrow cell expansion and functions as an effective immunosuppressant that prevents graft rejection in allogeneic transplantation models. - Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
Pre-aliquoted for Longevity: Solid-state aliquots offer significantly longer shelf life compared to stock solutions, ensuring maximum stability for every use.
