Nutlin-3a is a small-molecule MDM2 antagonist that inhibits the interaction between p53 and MDM2 with an IC50 of 90 nM in cell-free assays. By disrupting p53/MDM2 binding, Nutlin-3a stabilizes intracellular p53 and activates downstream p53-dependent signaling pathways. Consequently, this p53 activation leads to cell cycle arrest, autophagy, and apoptosis in p53-wild-type cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
