DAPT is a functional γ-secretase inhibitor that suppresses total Aβ production with an IC50 ofand Aβ42 levels with an IC50 of. By inhibiting γ-secretase, DAPT blocks Notch signaling pathways and decreases β-amyloid formation, leading to reduced Aβ40 and Aβ42 levels in neuronal cell models and in vivo tissues. Consequently, DAPT promotes neuronal cell differentiation and modulates cellular processes such as apoptosis and autophagy. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
