Adagrasib (MRTX849) is a potent, selective, and covalent KRASG12C inhibitor. It selectively modifies the mutant cysteine 12 residue in GDP-bound KRASG12C, locking the protein in its inactive state and blocking KRAS-dependent signaling. This suppression impairs downstream RAS/MAPK signaling, leading to decreased ERK phosphorylation, cell cycle arrest, and marked inhibition of cell proliferation in KRASG12C-mutant tumor models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
