Sitagliptin is a highly selective dipeptidyl peptidase-4 (DPP-4) inhibitor with an IC50 of 18 nM. By inhibiting DPP-4 enzymatic activity, Sitagliptin prevents the inactivation of incretin hormones such as glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). Elevated active incretin levels enhance glucose-dependent insulin secretion and suppress glucagon release, contributing to the regulation of glucose homeostasis in type 2 diabetes models. [1]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
