SAR405 is a highly selective low-molecular-mass kinase inhibitor of PIK3C3/Vps34 (Kd 1.5 nM) that demonstrates no significant activity against class I and class II PI3Ks or mTOR up to. By targeting Vps34 within the autophagy machinery, SAR405 prevents autophagosome formation and inhibits vesicle trafficking. Consequently, SAR405 synergizes with mTOR inhibitors to impair tumor cell proliferation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
