Biological Activity

RO4929097 is an orally bioavailable, potent small-molecule γ-secretase inhibitor that blocks the proteolytic processing and activation of Notch receptors. By inhibiting γ-secretase, RO4929097 prevents the cleavage and release of the intracellular Notch domain, downregulating expression of downstream transcriptional targets such as Hes1. This inhibition disrupts canonical Notch pathway signaling, ultimately suppressing tumor cell proliferation and promoting a less transformed, slower-growing cellular phenotype. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
