Clinical Trials

Several clinical trials are evaluating vociprotafib (RMC-4630) as monotherapy and in combination with ERK or KRAS G12C inhibitors for oncology indications, including advanced solid tumors, non-small cell lung cancer, colorectal cancer, pancreatic cancer, and KRAS-mutated malignancies. Spanning Phase I and Phase II studies, these trials are supported by industry sponsors (Revolution Medicines, Sanofi, Amgen) and institutional collaborators (The Netherlands Cancer Institute, Lustgarten Foundation). Current recruitment statuses across these studies encompass actively recruiting, active but not recruiting, completed, terminated, and unknown.

Compliance for Clinical Use

Mechanism and Biochemical Profile

Vociprotafib (RMC-4630) functions as a potent allosteric inhibitor of Src homology 2 domain-containing phosphatase 2 (SHP2), selectively binding the enzyme to block activation of the downstream RAS-RAF-MEK-ERK signaling cascade. This target pathway suppression impairs oncogenic signaling, inhibiting cancer cell proliferation and survival across clinically investigated malignancies such as KRAS-mutated solid tumors, non-small cell lung cancer, colorectal cancer, and pancreatic cancer.

Appendix RUO and cGMP Quality Standards

Quality Dimension RUO (Research Use Only) cGMP (Current Good Manufacturing Practice)
Clinical Applicability Prohibited in human clinical trials or medical diagnostics. Mandatory for human clinical trials (Phase I–III) and therapies.
Regulatory Status Non-regulated grade; exempt from drug manufacturing laws. Legally enforced by health authorities (e.g., FDA, EMA, NMPA).
Facility Environment Unclassified analytical or research laboratories. Validated Cleanrooms (ISO Class 5–8) with continuous monitoring.
Quality Control Basic purity and activity testing. Rigorous safety release testing (Sterility, Endotoxin, Mycoplasma).
Process Validation Basic equipment calibration; no process validation required. Full qualification (IQ/OQ/PQ) and complete batch records.
Quality Assurance Vendor self-declared without required formal QMS. Mandatory QA/QC unit, Change Control, CAPA, and vendor audits.
Regulatory Impact High risk of IND rejection if used as a critical raw material. Required for IND/NDA filings, supported by Drug Master Files (DMF).
