Biological Activity

PX-478 dihydrochloride is an orally active, selective hypoxia-inducible factor-1α (HIF-1α) inhibitor. It suppresses HIF-1α protein accumulation under hypoxic conditions without altering HIF-1α mRNA expression. Downstream, PX-478 decreases hypoxia-induced transcription of target genes such as VEGF. Consequently, the compound induces apoptosis, reduces tumor vascularization, and demonstrates potent anti-tumor activity in preclinical tumor xenograft models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
