Picropodophyllin (AXL1717) is a potent, selective inhibitor of IGF-1R (IC50 1 nM) that does not inhibit the phosphorylation of insulin receptor, FGFR, PDGFR, or EGFR. By blocking IGF-1R tyrosine kinase activity, it suppresses downstream AKT and MAPK signaling pathways. This inhibition impairs tumor cell proliferation, blocks cell cycle progression, and induces apoptosis, demonstrating strong antineoplastic activity across various cancer cell lines. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
