Midostaurin (PKC412) is a broad-spectrum, multi-targeted kinase inhibitor active against FLT3, c-Kit, PKC isoforms, PDGFRβ, and VEGFR1/2 with IC50 values ranging from 80 to. By inhibiting these kinases, it suppresses receptor autophosphorylation and disrupts downstream STAT5, AKT, and MAPK signaling pathways. This inhibition results in G1 phase cell cycle arrest and induces apoptosis in FLT3-mutated leukemia cell lines. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
