Biological Activity

Leptomycin B is an antifungal antibiotic that acts as a potent, covalent inhibitor of the nuclear export factor CRM1 (XPO1), inhibiting HIV-1 Rev nuclear export with an IC50 of. By alkylating a conserved cysteine residue in CRM1, it blocks NES-mediated nuclear export of target proteins. This disrupts nucleocytoplasmic transport, promoting nuclear retention of tumor suppressors and triggering cell cycle arrest and apoptosis in cancer cells. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
