Ceritinib selectively binds to the ATP-binding pocket of anaplastic lymphoma kinase (ALK) and related receptor tyrosine kinases including IGF-1R, InsR, STK22D, and FLT3, thereby blocking autophosphorylation and suppressing downstream RAS/RAF/MEK/ERK and PI3K/AKT/mTOR signaling pathways.
