Ceritinib (LDK378) is a potent, ATP-competitive inhibitor of ALK. By blocking ALK phosphorylation, it inhibits downstream signaling cascades, including the STAT3, AKT/mTOR, and ERK pathways. This target inhibition leads to suppressed cell proliferation and the induction of apoptosis in ALK-driven non-small cell lung cancer cells. Additionally, ceritinib exhibits significant antitumor efficacy in preclinical animal models, including crizotinib-resistant xenografts. [1][2]
