Biological Activity

Isavuconazole (BAL-4815) is an extended-spectrum triazole antifungal agent that inhibits cytochrome P450-dependent 14α-lanosterol demethylation (CYP51). By blocking CYP51, it disrupts ergosterol biosynthesis in fungal cell membranes, leading to depletion of ergosterol and accumulation of toxic methylated sterol precursors. This impairs membrane integrity and cellular viability, demonstrating broad-spectrum activity against yeasts, molds, and dimorphic fungi. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
