Biological Activity

LNP023 (Iptacopan) Hydrochloride is an orally bioavailable complement factor B inhibitor (IC=, Kd = that exhibits high selectivity over other proteases. By directly binding to factor B, it blocks the assembly of C3 and C5 convertases within the alternative complement pathway. This proximal pathway blockade suppresses membrane attack complex assembly, thereby preventing complement-mediated erythrocyte hemolysis and attenuating inflammatory tissue injury. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
