Ipatasertib (GDC-0068) is a potent, ATP-competitive pan-Akt inhibitor targeting Akt1 (IC50 5 nM), Akt2 (IC50 18 nM), and Akt3 (IC50 8 nM). It blocks the AKT/mTOR signaling pathway, suppressing downstream phosphorylation of PRAS40, S6, and eIF4G while promoting FOXO nuclear localization. This leads to selective inhibition of cell proliferation and robust antitumor efficacy in tumor xenograft models driven by activated PI3K/AKT signaling.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
