Mechanism and Biochemical Profile

Gossypol functions as an orally active polyphenol that binds anti-apoptotic Bcl-2 family proteins by blocking BH3 peptide interactions with Bcl-2 and Bcl-XL, while concurrently inhibiting 5α-reductase 1 and 3α-hydroxysteroid dehydrogenase. By suppressing anti-apoptotic signalling, gossypol reactivates intrinsic apoptotic pathways and suppresses cell proliferation, providing the mechanistic basis for its therapeutic investigation in solid tumors and hematologic malignancies such as non-small cell lung cancer and adrenocortical carcinoma.
