Gliclazide is a potent and selective blocker of pancreatic beta-cell ATP-sensitive potassium (KATP) channels via binding to sulfonylurea receptor subunits. Inhibition of KATP channels suppresses potassium efflux, triggering cell membrane depolarization and subsequent opening of voltage-dependent calcium channels. Elevated intracellular calcium levels promote calmodulin activation and insulin vesicle exocytosis, ultimately enhancing glucose-stimulated insulin release from pancreatic islets.

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
Pre-aliquoted for Longevity: Solid-state aliquots offer significantly longer shelf life compared to stock solutions, ensuring maximum stability for every use.
